Rapid single-tier serodiagnosis of Lyme disease

Earlier diagnosis can transform outcomes, but many powerful diagnostic technologies remain centralized, expensive, or difficult to deploy broadly. The Di Carlo Lab develops diagnostic platforms that combine microfluidics, cell biophysics, multiplexed assays, low-cost automation, and machine learning to move advanced measurements closer to the point of need.
One major example is deformability cytometry, a high-throughput method for measuring the physical properties of cells. This technology formed the foundation for Cytovale’s IntelliSep™ test, an FDA-cleared diagnostic for early sepsis detection. IntelliSep demonstrates how physical phenotyping of immune cells can provide rapid, clinically actionable information during acute disease.
The lab also develops vertical flow assays in collaboration with the Ozcan and Garner groups. These assays provide rapid, multiplexed biomarker detection in formats designed for point-of-care and resource-limited settings. Recent work demonstrated a rapid single-tier serodiagnostic test for Lyme disease using a paper-based vertical flow assay combined with machine learning.
